Whole Brain Imaging with STP Tomography

Serial Two-Photon (STP) Tomography is a proven, robust, imaging method that can generate fluorescent images of whole organs with sub-micron detail in the matter of hours. In this webinar Dr Tim Ragan reveals the technical underpinnings of STP Tomography, including tissue preparation, labeling, imaging and post process analysis, with a focus on neuroscience vascular applications.
Dr Berislav Zlokovic presents his study on using STP Tomography to generate a high-resolution 3-D model of the angioarchitecture in healthy and pericyte deficient mice. Recent studies have shown that pericytes regulate vascular stability and angioarchitecture. In the blood-brain barrier, pericytes degenerate in Alzheimer's Disease (AD), a neurodegenerative disorder characterized by early neurovascular dysfunction, elevation of amyloid β-peptide (Aβ), tau pathology and neuronal loss, leading to progressive cognitive decline and dementia. Based on the findings Dr Zlokovic’s group have designed a protocol that is practical and reproducible, and could become a new standard for neuropathological examination.
